The Zombie Cell Problem

In 2008, Judith Campisi's lab at the Buck Institute nailed down something cells do when stressed: instead of dying, they freeze in a damaged state called senescence. These cells stop dividing but refuse to clear out. They pump out inflammatory signals — the senescence-associated secretory phenotype, SASP — poisoning neighboring tissue. By your 70s, senescent cells have accumulated enough to measurably accelerate organ decline, fibrosis, and chronic inflammation.

First Kill Attempt (2015)

In 2015, James Kirkland and Jan van Deursen at Mayo Clinic proved the concept: genetically clearing senescent cells in mice extended median lifespan by 25% and compressed morbidity. The mice stayed healthier longer, not just alive longer. That paper in Nature landed like a grenade. It meant senescent cells weren't a side effect of aging — they were a driver. The field pivoted hard toward finding drugs that could do what the genetic trick did.
